The best way to describe Joker's personality is: inconsistent. At one moment he may be a harmless clown telling jokes to get a laugh, and in the very next moment he may just slit your throat with a playing card. He is completely unpredictable and there is no certain trigger that can be pinpointed that switches him from completely unconcerned to utterly devoted to your destruction.
The Joker's insanity appears to be a strange mixture of violently aggressive psychopathy, nihilism, and fixated narcissism. He has frequently been shown to have no regard for human life, and treats the pain and suffering of others as personal sport. It has been hypothesized by Arkham psychologists that the Joker actually exists in a state of almost "super-sanity". It is speculated that he redefines his own personality every day and can switch between personalities at the drop of a hat.
Another large part of Joker's personality is his obsession with Batman. He views the two of them as having a love/hate/murder relationship. Only Joker truly understands Batman and with their long history Batman is the only person to have come close to even slightly understanding the Joker. They're best buds, they're worst enemies, and neither of them would have it any other way - Joker knows its true.His obsession with the Dark Knight leads him to trying to outwit the man and also kill him quite often. Joker doesn't just want Batman dead though, its a game that they both play knowing that killing the other will not be a simple job and not one that anyone else will get to complete. Joker has saved Batman many times from death at the hands of another, and Batman has done the same. The really are the best of chums.

Unique Physiology: Due to his chemical submergence, Joker has gained beyond average advantages over normal humans as well as further exposure has tainted his physiology.
Pain Resistance: It is speculated that the Joker's exposure to chemicals may have heightened his resistance to pain. It is also implied that he is masochistic in nature, and thus enjoys any pain received, making it an ineffective way of dealing with him.
Tainted Blood: Joker's blood is tainted. It has been said that "being an avid consumer of his products, Joker's immunity to poisons has been built up over years of dedicated abuse." When a mosquito sucks the Joker's blood, it "writhes and whines, choking on tainted blood".
Joker Venom Immunity: The Joker is immune to his own venom as well as various similar toxins.
Cheating Death: The Joker has cheated death numerous times, even in seemingly inescapable and lethal situations. He has been, among other things, caught in explosions, electrocuted, shot repeatedly, and dropped from great heights, but he always returns to once again wreak havoc.
Cosmic Awareness: The Joker has, at times, shown to be fully aware of being a comic book character, even directly talking to the reader. However, most characters ignore this, due to the fact that he is criminally insane.
Indomitable Will: The Joker's insanity, or his frame of mind, leaves him immune to the Spectre's judgement, though warranted, as Batman explained once that the Joker doesn't actually know what is right and wrong, a crucial understanding the Spectre needs to judge. In a way, one has to know one's deeds are evil or wrong for the Spectre to judge, and the Joker doesn't. Also, both Jim Corrigan and Hal Jordan Spectres have had major problems with navigating the Joker's mind, Corrigan even managed to get lost in it and Jordan literally was blasted out of his boots to the moon when he tried.
Genius-level Intellect: The Joker is portrayed as highly intelligent and skilled in the fields of chemistry and engineering, as well an expert with explosives. From his first appearance onward, he has been consistently portrayed as capable of hijacking broadcasts - usually news programs - of both the television and radio varieties.
Inventive Skill: The Joker commits crimes with countless "comedic" weapons (such as razor-sharp playing cards, acid-spewing flowers, cyanide pies, and lethally electric joy buzzers) and Joker venom.

Hand-to-Hand Combat (Basic): The Joker has moderate skill in hand to hand combat. Over the years it has been shown that although Batman is stronger, the Joker is faster and more agile. The Joker has been known to be able to hold his own in hand-to-hand combat against Batman, however every time he is subdued by Batman, it is through physical force.
Intimidation: Joker is adept at torturing his victims endlessly, including shooting Batgirl, torturing Robin to death and beating Damian into submission.

His fascination with Batman; he can sometimes overplay his cards in an attempt to get Batman's attention and it is only because of his dependence on Batman that Joker always gets caught - but he doesn't mind that too much.

Any stories you may have heard of Joker's history prior to his transformation into the infamous Clown Prince of Crime are all rumor, most of them spread by the Joker himself. The man has no clear memory of his life before coming the homicidal maniac he is today and as far as Joker's concern he wasn't truly born until the day he fell into that vat of chemicals and came out stark white with green hair.

The Joker was in custody. How did he get there you ask? Why he simply walked in and checked himself in of course. "Ooh, cozy." Joker commented as he wiggled slightly in his new stylish straitjacket. "But hasn't anyone told you people white after labor day is a no no?" Joker paused for a moment, his bright red lips pressed tightly together in a thin red line, and then the irony hit him. "Ha ha ha ha!"
As much as Joker was amused the man standing before him in a white lab coat did not share his amusement. The psychiatrist clutched his clipboard tight to his chest with one hand as his other straightened the glasses on his face. "Why have you come here, Joker? I highly doubt it's to get yourself well. What is it you're up to?" The doc's words stung and if Joker wasn't strapped to some upright metallic bed-thing he felt he might have fallen over from the uncaring words. Ha. "Say, can't a guy swing by and - hee hee - visit old friends? Why feels like it was just yesterday when I jammed that number two pencil in your hand. Remember how you screamed like a little girl? Ha ha! Oh oh! Remember that other time I was cuffed and shackled to that chair and you got ♪tooo clooose♪ - and I bit half your ear off! Ah good times, good times. My especially favorite time was your birthday! When you thought I was drugged but I was just playing like I was drugged and then when you least expected it, bent you over the table and pulled down your pants and then I-"
"Joker!" The man was red in the face all of a sudden, his chest heaving as his heart pounded within his ribcage. Joker stifled his laughter as he looked down on the psychiatrist. "You always did have anxiety issues....and intimacy issues too! Hahahaha!" The entire board they'd strapped the clown to shook violently from his laughing tantrum. "Enough. You could have made this easier on yourself but of course you wouldn't. I know you have some scheme up your sleeve, but guess what? You've played this game a little too half cocked and you're not getting out of this situation. I'm going to leave out of this room and you will remain locked in, tied up in that jacket, and strapped to that slab. Congratulations, Joker, you're going to rot in this Asylum."
A slow smile crept to the doctor's face, a glint shining in his eye as he revelled in his victory over the clown. "Ha ha ho ho ho. I came here of my own free will, doc. Do you really think I'd come if it weren't for a specific reason? And do you really think an egghead dips&!t like you can change anything I've already put into play? That's cute...but stupid!" The Joker's voice dripped with malice, no longer joking and having a good time with an old friend. "Now ask me again why I'm here." As he instructed so the psychiatrist did. "I'm here for you, silly. I just haaad to be here in person to see your face...after you called your daughter."
The man's face lost color, his heart missed a beat, and his stomach crashed to his feet. "M-m-my daughter? Wh...what have you done!?" Joker now had a balding, bifocalled man clinging to the front of him. The doc totally clash with his snazzy new jacket. "Why don't you give her a ring-a-ding?" His clipboard dropped to the tiled floor and the man quickly fumbled with his pockets to retrieve his cell phone, all the while Joker watched him closely with a giant smile plastered on his face.
The phone rang. It rang again. And then finally it picked up. "Jessica? Jessica are you there?!" The doc clutched his phone like a life line and Joker's smile stretched even wider. "Daddy? Noooo- BOOOOOM! The explosion over the line was so loud the psychiatrist had to pull his phone away from his ear. When the phone was pressed back against his face the line was already dead - much like his daughter. "Jessica? Jessica!"
Joker watched the scene unfold and shifted his face into an expression of mock pity. "Oh dear. Did I forget to mention her phone was the detonator to the explosives I had strapped to young Jessica and a call from your phone would trigger said detonator? Whoops must have slipped my mind!" The Joker cackled madly as his long time friend dropped down to his knees, cell phone still clutched in hand, and began to sob his poor little eyes out. "HAAHAHAHAHAHA!"
